摘 要: | 目的]鉴定引起病死鹅肝脏表面出现大量针尖大、白色坏死点的致病菌。方法]取肝涂片镜检,然后进行瑞氏-吉姆萨染色。用普通肉汤培养基和鲜血培养基进行分离和纯化,然后用巴氏杆菌特异性引物进行PCR扩增,将扩增产物克隆至PMD18-T载体上,送至生物公司进行测序。结果]序列同源性分析表明,分离菌与多杀性禽巴氏杆菌的相似性为99%,结合生化试验结果确定其为多杀性巴氏杆菌。结论]研究可为鹅巴氏杆菌病的防治提供依据。

Isolation and Identification of Pasteurella multocida in Goose |

Abstract: | Objective] To identify the pathogen that caused a large number of white necrosis of the needle point in the liver surface of dead geese.Method] The microscopic examination of liver smear was observed,and Wright-Giemsa staining was conducted.After using ordinary broth and blood culture medium for separation and purification,PCR amplification was carried out by Pasteurella multocida specific primers.The purified product was cloned into PMD18-T vector,then sent to the biological company for sequencing.Result]The similarity of the obtained sequencing result by comparison to Pasteurella multocida was 99%,which was determined as Pasteurella multocida combined with biochemical test results.Conclusion] The research could provide basis for the prevention and control of pasteurellosis. |
