茶新菇液体发酵培养基优化研究

摘    要:目的]为了获得培养茶新菇的最优碳源、氮源、无机盐配比和最适培养条件。方法]通过对茶新菇菌的液体深层发酵培养基组分中的碳源、氮源、无机盐的种类进行选择性试验分析,寻找茶新菇液体发酵培养基的最佳组合。结果]单因子试验证明最优碳源为蔗糖、最优氮源为蛋白胨、最优无机盐为KH2PO4;其中碳源浓度对菌丝体产量的影响最大,其次是氮源,最小是无机盐,组分配方的最佳组合是:蔗糖3%、蛋白胨1%、KH2PO40.10%,最适培养条件为25℃、转速150 r/min、接种量5%、摇床振荡培养4 d。结论]在最优组分配比和最适培养条件下,茶薪菇菌丝体生物量最高,该研究为其高效生产提供了定量的理论数据。

Abstract:Objective]The purpose of research was to obtain the optimum component matching of carbon source,nitrogen sources and inorganic salts ratio and optimum cultivation conditions for culturing Agrocybe aegerita.Method]Selecting tests were carried out for the kinds of carbon sources,nitrogen sources and inorganic salts of liquid fermentation medium component in Agrocybe aegerita to study the optimum combination of liquid fermentation medium in Agrocybe aegerita.Result] Single-factor test proved that the optimum carbon sources was sucrose,the optimum nitrogen sources was peptone and the optimum inorganic matters was KH2PO4.The concentration of carbon sources had the greatest effect on mycelium yield,followed by nitrogen sources,and the lowest was inorganic matters.The optimum ingredient matching in liquid fermentation medium was as following: 3% sucrose,1% peptone and 0.10% KH2PO4.The optimum culture conditions was as following: temperature of 25 ℃,revolution speed of 150 r/min,inoculation of 5% and oscillating culture on shaking table for 4 days.Conclusion] The mycelium biomass of Agrocybe aegerita was the highest under the optimum component matching and optimum culture condition.The research provided quantitative theoretical data for efficient production of Agrocybe aegerita.
