不同浓度EMS对苦荞种子发芽的影响

摘    要:目的]确定荞麦种子发芽的适宜甲基磺酸乙酯(EMS)诱变剂量,为进一步获得荞麦优良性状并培育符合EMS诱变育种需求的新品种打下坚实的基础.方法]以山西省当地的苦荞Fagopyrum tataricum(L.)Gaertn]品种晋荞麦4号种子为材料,用不同浓度梯度(0.3%、0.5%、0.7%、1.0%、1.5%、1.7%)EMS诱变处理不同时间(4、8、12 h),分析苦荞种子发芽率和发芽势.结果]随着EMS诱变剂浓度的增加,晋荞麦4号种子发芽势、相对发芽势和发芽率、相对发芽率均呈下降趋势,当EMS浓度为0.3% ~1.0%时与对照没有显著差异,当EMS浓度为1.5%和1.7%时与对照差异极显著.EMS处理4、8h对晋荞麦4号种子发芽率、相对发芽率都没有大的影响,当处理时间为12 h时,与对照有显著差异;对于发芽势和相对发芽势,这3个处理时间没有显著差异.结论]EMS诱变苦养的半致死的适宜浓度为1.7%,适宜的处理时间为12 h.

Effects of Different Concentration EMS on Germination of Tarary Buckwheat[Fagopyrum tataricum (L.) Gaertn] Seeds

Abstract: Objective ] The aim was to confirm on the suitable EMS dosage of tarary buckwheat seed gemination,and further lay a solid founda- tion for obtaining excellent traits of buckwheat and breeding the new varieties meeting EMS mutagenesis breeding. Method] The seeds of Jinqiao- mai No. 4 ,a Shanxi local tarary buckwheat variety,were mutagenized by different concentration(0.3% ,0.5% ,0. 7% ,1.0% ,1.5% ,1.7% ) EMS for 4,8 and 12 h,to analyze their gemination rate and gemination vigor. Result ] With the increase of EMS mutagenic agent concentration, the gemination vigor,relative gemination vigor, gemination rate and relative gemination rate of Jinqiaomai No. 4 all showed the decrease trend, they had no significant difference with control when EMS concentration was 0.3% - 1.0%, while had very significant difference with control when EMS concentration was 1.5% and 1.7%. Using EMS mutagenic agent to mutagenize Jinqiaomai No.4 seeds for4 and 8 h had no big effects on its gemination rate and relative gemination rate,while when the treatment time was 12 h,they had significant difference with control; For gemination vigor and relative gemination vigor,this three treatment time had no significant difference. Conclusion] The suitable concentration and time of the median lethal dose of EMS mutagenizing tarary buckwheat seed is 1.7% and 12 h,resp.
