洋葱核型分析及有关制片方法的探讨

目的]研究洋葱核型并探索其较佳的制片方法，为进一步研究洋葱遗传改良奠定基础。方法]进行洋葱核型分析，其根尖细胞制片技术则是关键。研究比较了不同的预处理时间（1．5、2．0、3．0h）、不同固定时间（18．0、21．0、24．0h）、不同的染液（醋酸洋红染液、碱性品红染液）对洋葱根尖细胞染色体制片效果的影响。结果]结果发现，洋葱根尖的较好的预处理时间为3．0h，较好的染液为碱性品红染液；在此基础上，通过洋葱根尖细胞染色体核型分析，发现试验所用洋葱的染色体数为2n=2x=16。在洋葱的16条染色体中没有发现具有随体的染色体，但发现有端部着丝点染色体；经过对染色体的测量与计算，该研究中洋葱的核型公式为2n=2x=16=14m＋2t，核型不对称系数为58．2％。结论]这种核型公式及核型不对称系数对洋葱品种资源的鉴定具有一定的参考价值。
