金钗石斛多糖对脂多糖诱导的小鼠腹腔巨噬细胞分泌TNF-α·NO的影响

摘    要:目的]研究金钗石斛多糖对脂多糖(LPS)诱导的小鼠腹腔巨噬细胞分泌肿瘤坏死因子-α(TNF-α)、一氧化氯(NO)的影响。方法]用不同浓度金钗石斛多糖作用于小鼠腹腔巨噬细胞,LPS作为诱导剂,采用放射免疫法检测细胞培养液中TNF—α的含量,硝酸酶还原法测定细胞培养液中NO的含量,荧光法测定细胞内诱导型一氧化氮合酶(iNOS)的活性,实时PCR(real—time PCR)法测定TNF-α mRNA、iNOS mRNA的表达。结果]与LPS组比较,金钗石斛多糖在50~400mg/L范围内可干预LPS对小鼠巨噬细胞的作用,使小鼠腹腔巨噬细胞TNF—α、NO合成减少,iNOS活性降低。TNF-α mRNA、iNOS mRNA的表达降低。结论]金钗石斛多糖可改善LPS对小鼠腹腔巨噬细胞的作用,使TNF—α mRNA、iNOS mRNA的表达降低,TNF—α、NO合成减少,从而起到抗炎的作用、

Abstract:Objective]Study on the effects of Dendribum nobile polysaccharide on tumor necrosis factor-α(TNF-α) and nitric oxide(NO) release by mouse peritoneal macrophage induced Lipopolysaccharide(LPS) in vitro.Method]The peritoneal macrophages of mice were conducted with LPS in the presence of Dendribum nobile polysaccharide at various concentration.TNF-αwas detected by radioimmunoassay,NO content of cell culture fluid was determined by nitrate reductase method,the activity of inducible nitric oxide synthase(iNOS) was measured by fluorimetry assay.The expression of TNF-αand iNOS mRNA were observed by real-time polymerase chain reaction(real-time PCR),Result]Compared with the control group,Polysaccharides of Dendribum nobile could markedly inhibit the secretion of TNF-αand NO and decrease the activity of iNOS and down-regulate the expression of TNF-αm RNA and iNOS mRNA in a dose dependent manner.Conclusion]Dendribum nobile polysaccharide can improve TNF-αmRNA、iNOS mRNA and its protein expression induced by Lipopolysaccharide in mouse peritoneal macrophage that may play a very important role in the prevention of inflammation.
