嗜热纤维素分解菌TH3-9诱变选育及产酶条件初步研究

摘    要:以嗜热侧孢霉(Thermophilic sporotrichum)为出发菌株进行紫外诱变,选育出TH3-9突变株。该菌株的CMCase和FPAase分别提高到原始菌株的2.6和2.0倍。液体培养产酶条件及正交试验研究结果表明,最适碳源为稻草粉+甘蔗渣(总含量为2.0%时,稻草粉∶甘蔗渣为1∶4),最适氮源是蛋白胨(0.4%),KH2PO4最适含量为0.3%,MgSO4.7H2O最适含量为0.03%,最适pH值为5.5,最适培养温度为45℃,培养第2天FPAase最高,培养第4天CMCase最高。采用优化配方,在45℃下培养,测得CMCase为86.8 IU,FPAase为22.6 IU。

Abstract:Thermophilic sporotrichum was mutagenized with the ultraviolet rays and the strain TH3-9 was attained.CMCase production of the strain TH3-9 was increased by 2.6 times and FPAase production was increased by 2.0 times,compared with the parent strain.The optimum condition of liquid fermentation for cellulase production of TH3-9 was explored.It was found that the optimum carbon source was bagasse+straw powder(total content was(2.0%) and the proportion of them was 1∶4),the optimum nitrogen source was peptone(total content of it was 0.4%) and the optimum culture temperature was 45?℃.The optimum content of KH_2PO_4 was 0.3%,the optimum content of MgSO_4·7H_2O was 0.03% and the optimum pH was 5.5.When cultured under these conditions,CMCase was 86.8?IU and FPAase was 22.6?IU.
