转基因玉米根际土壤DNA的提取

摘    要:目的]探索转基因玉米根际土壤DNA提取的方法。方法]采用直接裂解方式,通过玻璃珠、溶菌酶和SDS共同作用于转基因玉米MON863和Bt 176的根际土壤中,提取微生物DNA。结果]该方法能从两种转基因玉米根际土壤中提取到20 kb的完整DNA片段。结论]所得DNA完全适用于PCR扩增和酶切的要求。

Abstract:The research aimed to screen a kind of extraction method for genomic DNA in rhizosphere soil grown with transgenic corn.A direct lysis manner for extracting DNA directly from rhizosphere soil of transgenic corn——MON863 and Bt 176 was designed with glass bead-lysozyme-SDS method.The results showed that intact DNA fragments over 20 kb were gained from rhizosphere soil planted with two cultivars of transgenic corn.DNA extracted suited to PCR amplification and RFLP.
