摘    要:目的]对胀果甘草愈伤组织进行干旱胁迫处理,探讨干旱胁迫对愈伤组织的生长、生理指标和甘草黄酮含量的影响。方法]采用不同质量分数(0、15%、20%、25%、30%)的PEG-6000模拟干旱胁迫对胀果甘草愈伤组织进行胁迫处理,测定其生长量,MDA、POD以及黄酮含量。结果]随着胁迫程度的增加,甘草愈伤组织的生长量受到抑制,MDA含量呈上升趋势,POD含量呈现先上升后下降的趋势。25%PEG-6000处理子叶愈伤组织时甘草黄酮含量为4.80mg/g,约为对照的1.379倍。下胚轴愈伤组织经过干旱胁迫后甘草黄酮含量有所下降。结论]一定浓度的干旱胁迫可以提高子叶愈伤组织的甘草黄酮含量。

Effect of Drought Stress on Total Flavonoids Content in Glycyrrhiza inflata Callus

Abstract:Objective] The effect of PEG-6000 simulated drought stress on the growth of callus,physiological indicators and the content of licorice flavonoids in callus were studied.Method] With different mass fraction (0,15%,20%,25%,30%) of PEG-6000 simulated drought stress on Glycyrrhiza inflata callus,the growth amount,MDA,POD and flavonoid content of the callus were measured.Result] With the stress level increased,the amount of callus growth showed a downward trend,MDA on the rise,POD generally showed up first,then downward trend.The licorice flavonoids content of cotyledon callus increased to 4.8 mg/g under the 25% PEG-6000 treatment,it was about 1.379 times of the control.Licorice flavonoid content of hypocotyl calli decreased under the PEG simulated drought stress.Conclusion] The certain drought stress could increase the licorice flavonoids content in cotyledon callus of Glycyrrhiza inflata.
