红三叶寡肽对小鼠免疫功能的影响

摘    要:目的]以由红三叶草叶蛋白制备的寡肽为原料,研究其对小鼠生长及免疫功能的影响。方法]选用64只,体重18~24g的昆明小鼠,随机分成4组,每个处理16只,雌雄各半,分笼饲养。所有小鼠每天上午灌胃一次,灌胃量为0.3ml,其中,3个试验组寡肽溶液灌胃浓度分别为5.0、1.0、0.1mg/ml,对照组灌胃生理盐水,试验期21d。结果]①灌胃0.1mg/ml寡肽试验组体增重极显著地高于对照组(P<0.01),灌胃1.0mg/ml寡肽试验组增重显著高于对照组(P<0.05);②灌胃0.1mg/ml寡肽试验组的脾重系数极显著高于对照组(P<0.01),所有试验组的胸腺重系数均显著提高(P<0.05),其中,5.0mg/ml试验组与对照组间差异达到极显著水平(P<0.01);③1.0mg/ml试验组的溶血素水平极显著高于对照组(P<0.01),而0.1mg/ml试验组与对照组相比也达到了显著水平(P<0.05);④0.1mg/ml试验组小鼠右耳增厚程度显著高于对照组(P<0.05),表明小鼠迟发性变态反应明显;⑤各试验腹腔巨噬细胞吞噬鸡红细胞的吞噬百分率均显著提高(P<0.05),其中,1.0、0.1mg/ml试验组的影响达极显著水平(P<0.01);⑥1.0mg/ml试验组T淋巴细胞百分比显著高于对照组(P<0.05)。结论]红三叶草寡肽能够有效促进小鼠的生长及免疫力的提高,且以寡肽溶液灌胃浓度为1.0、0.1mg/ml时的效果更为明显。

Abstract:Objective The effect of oligopeptide extracted from Trifolium Pratense on growth and immune functions of mouse was studied.Method Sixty-four Kunming mice with body weights between 18 g and 25 g were divided into 4 groups and randomly assigned to 4 treatments of sixteen mice for each(eight males and eight females).Males and females were fed in separate cages.Three groups were supplementded with 5.0 mg/ml,1.0 mg/ml and 0.1 mg/ml oligopeptide solution respectively and one group was given normal saline as control.The solution volume for each mouse was 0.3 ml everyday.The experiment lasted for 21 days.Result Supplementation of oligopeptide increased weight gain(P<0.05 and P<0.01 for 1.0 mg/ml and 0.1 mg/ml groups respectively).The spleen weight of 0.1 mg/ml group was significantly higher than the control(P<0.01).All experimental groups had higher thymus gland weight(P<0.01 for 5.0 mg/ml group and P<0.05 for the rest).Supplementation of oligopeptide increased hemolysin(P<0.01 and P<0.05 for 1.0 mg/ml and 0.1 mg/ml groups respectively).Right ear thickness of 0.1 mg/ml group increased(P<0.05),indicating obvious delayed type hypersensitivity.The percentage of macrophage devouring chicken red blood cell increased(P<0.01 for 1.0 mg/ml and 0.1 mg/ml groups and P<0.05 for the rest).1.0 mg/ml group had a higher percentage of T lymphocyte than the control(P<0.05).Conclusion Oligopeptide extracted from Trifolium Pratense could improve some immune functions of mouse.The oligopeptide solution at 1.0 mg/ml and 0.1 mg/ml with a volume of 0.3 ml everyday had much significant result.
