摘    要:目的]探讨抗稻瘟病基因Pib4的转基因。方法]利用PCR法克隆Pib基因的NBS功能结构域到双元载体pCAMBIA1301上,以粳稻品种农大18的成熟胚诱导出愈伤组织,以愈伤组织作为转化的受体,采用农杆菌介导法转化Pib4基因。结果]经PCR和GUS活性检测表明外源基因已整合到水稻基因组中并获得表达;用稻瘟病菌接种阳性植株,获得了抗稻瘟病菌的水稻植株。结论]为合理利用抗病基因和育种提供了参考。

Transgene of Rice Blast Resistance Gene Pib4

Abstract:Objective] The aim was to discuss the transgene of rice blast resistance gene Pib4.Method] NBS domain of Pib4 gene was amplified by PCR and subcloned into the binary vector pCAMBIA1301,the Pib4 gene by Agrobacterium tumefaciens were transferred into calli which derived from mature embryos of japonica rice Nongda 18.Result] Transgenic rice was confirmed by PCR and GUS analysis.Positive transgenic plants were inoculated with Magnaperthe grisea fungus,which had the same resistance with control plant.Conclusion] All those provide references for using disease-resistant gene and breeding rationally.
