基于实验条件下提取和测定麻黄碱方法的选优研究

目的]确定实验条件下索氏法提取麻黄碱和酸性染料比色法测定含量的最佳参数。方法]以不同生长期的麻黄草为材料,干燥粉碎后,用索氏法提取麻黄碱,酸性染料比色法测定含量。研究了不同波长、pH值,溴百里酚蓝用量对麻黄碱提取效果的影响。结果]麻黄碱溶液与pH值为6.8的溴百里酚蓝缓冲溶液混合后,在410 nm处有最大吸收峰 麻黄碱溶液与不同pH值的溴百里酚蓝缓冲溶液混合后,与pH值为6.8的缓冲溶液混合后的吸收峰最大 在pH值为6.8溴百里酚蓝缓冲溶液中,麻黄碱与溴百里酚蓝的体积比超过1∶4时,在410 nm处的吸光度基本保持恒定。结论]酸性染料比色法的最低检出限为10 mg/L,平均回收率为98.39%。
