淡紫拟青霉几丁质酶提取工艺的优化研究

摘    要:目的]为优化淡紫拟青霉几丁质酶的提取工艺.方法]以硫酸铵溶液为提取剂,先通过单因素试验,确定硫酸铵浓度、透析温度、透析时间和透析液酸碱度的最佳水平,然后通过正交试验确定最住浸提条件.结果]淡紫拟青霉几丁质酶的最佳提取工艺为:硫酸铵浓度60%、透析温度30 ℃、透析时间2d,透析液pH 6,在该条件下几丁质酶酶活可达0.0534 U.结论]该结果可以为几丁质酶的进一步开发和工业化生产提供一定科学依据.

Abstract: Objective] The aim was to optimize the extraction process of Paecilomyces lilacinus chitinase. Method] With ammonium sulfate solution as extraction agent, firstly the best levels of ammonium sulfate concentration, extracting temperature, extracting time and extracting pH were detemined through single factor test. And the best extraction condition was determined by orthogonal experimental design. Result] The best extraction condition for chitinase of Paecilomyces lilacinus was as follows : ammonium sulfate concentration 60% , extracting temperature 30 ℃, leaching time two days and leaching pH 6. Under this condition, the chitinase activity reached 0.053 4 U. Conclusion] The results provided a scientific basis for the further development and industrialized production of chitinase.
