地黄叶片提取物抗氧化活性及其薄层色谱的初步研究

摘    要:目的]为开发利用地黄资源提供依据。方法]以新鲜地黄叶片为试材,将其热烫、打浆、过滤、真空浓缩干燥后,分别用乙醇、丙酮、氯仿、乙醚、石油醚提取浓缩物中抗氧化活性物质,比较不同溶剂提取物的抗氧化作用。并用化学预示法和薄层层析法对抗氧化活性物质的成分进行定性分析。结果]5种有机溶剂的提取物均具有抗氧化作用,其中,乙醇提取物的抗氧化作用最强,其最适用量为0.14%。0.14%乙醇提取物的抗氧化活性略低于0.02%的茶多酚,但高于0.02%的BHT和维生素C。化学预示和薄层色谱分析显示,地黄叶片的抗氧化活性物质主要为黄酮、萜类、有机酸和酚类化合物。结论]确定了地黄叶片有机活性物质的组合及最佳提取溶剂。

On the Antioxidation Activity with Thin Layer Chromatography of the Extraction from Rehmannia glutinosa Libosch leaves

Abstract:Objective] To provide basis to develop and utilize Rehmannia glutinosa Libosch.Method] The fresh Rehmannia glutinosa Libosch were treated with hot water,beaten,filtrated,dried in the vacuum condition and then antioxidation materials was extracted from the concentrated stuffs by ethanol,acetone,chloroform,ether and petroleum benzene and their antioxidation activity was compared.The components of antioxidation materials were analyzed quantitatively by chemical predictive method and thin layer chromatography.Result] All extracts from 5 kinds of organic solvents has an antioxidation effect,among which ethanol extract has much stronger antixoidation performance with an optimum concentration of 0.14%,which is slightly lower than that of 0.02% Tea Polyphenols but higher than that of 0.02% BHT and Vitamin C.The analysis result showed that the main components of antioxidation materials from Rehmannia glutinosa Libosch leaves were flavonoids,terpenols,organic acid and phenol compounds.Conclusion] This study figured out the components of organic active materials from Rehmannia glutinosa Libosch leaves and the optimal extracting solvents.
