摘    要:目的]开发高纯度费菜总黄酮的分离纯化工艺。方法]以大孔树脂为载体,对上样量及层析条件进行优化,再进一步采用结晶的方法进行纯化。结果]选用聚合物纳米微球PS RPC-300作为最佳层析填料,洗脱速度为5 m L/min,洗脱剂为70%乙醇,上样量为5.0 g/kg填料,结晶溶剂为丙酮;在此优化条件下所得产品的纯度为95.1%。结论]此分离纯化方法简便可靠,分离效果好。

Study on the Separation and Purification of Total Flavonoids from Sedum aizoon L.
Abstract:Objective] The research aimed to develop the separation and purification processes of high-purity total flavonoids from Sedum aizoon L.Method]With the macroporous resin as the carrier,the loading volume and the chromatographic conditions were optimized,and then further purified by crystallization method.Result]Type of PS RPC-300 nanoscale polymer particles was selected as the best chromatography filler.The optimal conditions were as follows:the elution speed was 5.0 mL/min,70% ethanol was used as the eluant,the amount of sample was 5 g/kg filter,and acetone was used as the crystallization solvent.The purity of the final product was 95.1% under the optimal conditions.Conclusion]The separation and purification method is simple and reliable,and the separation effect is good.
