摘    要:目的]为乌药的规模化人工育苗提供理论依据。方法]以乌药植物当年生健壮枝条为外植体,研究不同激素种类及浓度对其不定芽的诱导及生根的影响,筛选出最佳培养基。结果]适宜于乌药组织培养的最佳外植体取材时间为6~7月,以当年生带芽茎段的嫩枝为最适宜;2.5 mg/L 6-BA是乌药不定芽增殖时最有效的浓度;诱导乌药外植体不定芽增殖的最佳培养基为MS+6-BA 2.5mg/L+NAA 0.2 mg/L;乌药苗在增殖过程中的继代周期以40~50 d为宜;最佳壮苗培养基为MS+6-BA 1.0 mg/L+NAA 0.2 mg/L;最佳生根培养基为MS+NAA 0.5 mg/L,生根率98%~100%。结论]采用合适的外植体以及激素种类与浓度为最佳配比的培养基可使传统的中药材乌药植物成功地诱导不定芽的分化、生根与再生。

Abstract: Objective] The study aimed to provide theoretical basis for the large-scaled artificial breeding of Lindera aggregate(Sims)Kosterm seedlings.Method] In tissue culture the current-year hale branches of L.aggregate were taken as explants to research the effects of different hormones at different concn.on the inducing and rooting of adventitious buds of L.aggregate,and select the optimum medium.Result] The optimum time for selecting the explants that fitted for the tissue culture of L.aggregate was from June to July,and the current year's twig stem segment with bud was better.The most effective concn.of 6-BA for the adventitious bud multiplication of L.aggregate was 2.5 mg/L.The optimum medium for the multiplication with adventitious buds of L.aggregate was MS + 6-BA 2.5 mg/L+ NAA 0.2 mg/L,The appropriate subculture cycle was 40-50 d in multiplication culture of L.aggregate seedlings.The optimum medium for strengthening shoots was MS + 6-BA 1.0 mg/L + NAA 0.2 mg/L and that for shoot rooting was MS + NAA 0.5 mg/L,with the rooting rate of 98%~100%.Conclusion] Adventitious buds differentiation,rooting and regeneration of the traditional Chinese medicines L.aggregate were successfully induced by using suitable explants and optimizing the mediums with different hormones and concn.
