摘 要: | 目的]探讨中华鳖和杂交鳢血清免疫球蛋白的同源性及其含量。方法]采集中华鳖和杂交鳢血清与25种商品二抗共同抚育,进行免疫印迹及酶联免疫分析。结果]中华鳖和杂交鳢血清可与兔抗鸡IgM、兔抗猪IgG、兔抗鸡IgG、兔抗大鼠IgM、兔抗猴IgG和羊抗人IgG等6种二抗发生免疫印迹反应。利用鸡IgG、鸡IgM、猪IgG、大鼠IgM、猴IgG和人IgG的ELISA试剂盒测定了中华鳖和杂交鳢血清免疫球蛋白水平,中华鳖血清免疫球蛋白浓度为28.5~215.4μg/L,杂交鳢血清免疫球蛋白浓度为24.5~201.5μg/L。结论]该研究能够丰富中华鳖、杂交鳢特异性免疫系统的研究,对中华鳖、杂交鳢的健康养殖有一定的指导意义。

Analysis of Immunoglobulin in the Serum of Trionyx sinensis and Ophiocephalus argus Cantor(♂)×Ophiocephalus maculatus(♀) by Western blot and ELISA |

Abstract: | LIU Kai et al ( Hangzhou Academy of Agricultural Sciences, Hangzhou, Zhejiang 310024) Abstract Objective] The research aimed to study the homology and content of serum immunoglobulin in Trionyx sinensis and Ophiocephalus argus Cantor ( ♂ ) × Ophiocephalus rnaculatus( ♀ ). Method] The serum of T. sinensis and O. argus Cantor ( ♂ ) × 0. rnaculatus( ♀ ) was sampled to cultivate with 25 commodities secondary antibodies for Western blot and ELISA analysis. Result] The serum of T. sinensis and O. argus Cantor (♂) × 0. maculatus( ♀ ) reacted with six kinds of secondary antibodies(rabbit anti-chicken IgM, rabbit anti-pig IgG, rabbit anti-chicken IgG, rabbit anti-rat IgM, rabbit anti-monkey IgG and goat anti-human IgG)for Westem blot. The serum immunnoglobulin level of T. sinensis and O. argus Cantor (♂)× 0. maculatus( ♀ ) was determined by using ELISA kits of chicken IgG,chicken IgM,swine IgG,rat IgM ,mongkey IgG and human IgG. The serum immunnoglobulin concentrations of T. sinensis and O. argus Cantor (♂) × 0. maculatus( ♀ ) were 28.5 -215.4 and 24.5 - 201.5 μg/L respectively. Conclusion ] This study could enrich the specific immune system research of T. sinensis and O. argus Cantor ( ♂ ) × 0. maculatus( ♀ ) and it was of certain guiding significance for the healthy breeding of T. sinensis and 0. argus Cantor (♂) × O. maculatus ( ♀ ). |
