烟叶中性挥发性成分的GC和GC-MS分析

摘    要:为了选择中性挥发性成分提取方法和灵敏度高的检测仪器,确定仪器检测的条件。采用顶空共蒸馏法对烟叶的中性挥发性成分进行提取和分离,用气相色谱仪(GC)和色谱-质谱联用仪(GC-MS)进行检测,提取并鉴定了数十种烟叶中性挥发性成分,并对每种物质计算了相对标准偏差,同时利用已有的标准物质,进行了方法的准确性评价。确定了色谱与质谱条件,即进样口的温度为280℃,进样针条件为注入样品后,先停留5 s再拔针,程序升温的初始温度为40℃,检测器的开始检测时间为2.5 min(温度为40℃),传输线和离子源的温度分别为250和200℃,样品检测质量数范围在300以下。分析出烟草中57种主要中性挥发性成分的相对标准偏差和含量以及35种标样的回收率。确定、优化了仪器检测条件。

Abstract:The research was conducted to select the methods of extracting neutral volatility component and the detecting instruments with high sensitivity in order to ascertain the detection conditions of instruments.Neutral volatility components were distilled and separated by headspace co-distillation method,and then detected with GC and GC-MS.Dozens of neutral volatility components were distilled and identified,and the relative standard deviations(RSD) for each matter were calculated and the method veracity was evaluated by using existing standard matter.The conditions in gas chromatography and mass spectrometry were confirmed as follows: the injection port temperature was 280 ℃;the injection needle should first remain for 5 s and then pull out after injecting samples;the initial temperature for programmed temperature was 40 ℃;the beginning detection time for the detector was 2.5 min at the temperature of 40 ℃;the temperatures for transmission line and the ion source were 250 ℃ and 200 ℃ respectively and the range of mass numbers in sample detection was below 300.The RSD and content of 57 major neutral volatility components in tobacco and the recovery rates of 35 standard samples were analyzed.The detection conditions of instruments were determined and optimized.
