雷蘑液体发酵工艺优化研究

摘    要:在雷蘑液体发酵培养基优化的基础上,研究了适于雷蘑深层发酵的初始pH值、接种量、装液量、发酵温度和时间及摇床转速对菌丝生长和胞外多糖产量的影响。并通过L16(45)正交试验对发酵工艺进行了优化,结果表明:在初始pH值6.5,接种量10%,装液量100ml/500 ml,培养温度28℃,培养时间9 d,转速140 r/min的发酵条件下,所得胞外多糖量为93.61 mg/100 ml。

Abstract:The effect of initial pH,volume of medium,amount of inoculation,temperature,time and on the growth of mycelium and production of exopolysaccharide was studied based on the optimal fermentation medium in submerged cultivation with Clitocybe gigantean AS 5.105.The orthogonal test of L_(16)(4~5) showed the maximum yield of EPC was obtained under the circumstance of volume of medium: 100 ml/500 ml;the amount of inoculation: 10%;pH value: 6.5;rotate speed: 140 r/min;temperature: 28 ℃ and time: 9 days.The maximum yield of EPC was 93.61 mg/100 ml.
