高产漆酶菌株Bacillus sp.CLb的筛选及其对染料脱色效果的研究

摘    要:目的]为了筛选出一株高产漆酶的菌株。方法]利用含铜的富集培养基从土壤中筛选出漆酶活性较高的菌株,结合形态学、生理生化特性及16S rDNA序列分析对菌株的分类地位进行鉴定,研究菌株的生长特性及菌株的芽孢漆酶对常用染料的脱色效果。结果]该菌株属于芽孢杆菌属,命名为Bacillus sp.CLb。菌株CLb最适生长温度为37℃,最适生长pH为7.0,菌株能在含1 mmol/L Cu2+的培养基中生长,具有很强的耐铜性。以丁香醛连氮为底物,测定其芽孢漆酶活性,漆酶活性高达46.1 U/g干重。菌株CLb芽孢漆酶的最适pH为7.0。在介体乙酰丁香酮存在的脱色体系中,菌株CLb芽孢漆酶在4 h内对活性黑以及在2 h内对靛红的脱色率均达到93.0%,在6 h内对活性亮蓝和结晶紫脱色率分别为79.0%和92.5%。结论]菌株CLb芽孢漆酶在介体乙酰丁香酮存在的体系中对常用染料具有很高的脱色率。

Abstract:Objective] The research aimed to screen a strain with high laccase activity.Method] Enrichment medium added with copper ions was used to screen bacteria with laccase activity from forest soil.A bacterium highly producing-laccase was identified by morphological,physiological and biochemical characteristics with 16S rDNA sequence analysis.The growth characteristics of the strain and the decolorization effect to dyc were researched.Result] The strain was identified as Bacillus sp.CLb.The optimum growth temperature of strain CLb was 37℃ and the optimum growth pH was 7.0.The strain CLb could grow in culture medium containing 1 mmol/L Cu2+.Its spore laccase activity was 46.1 U/g dry weight used syringaldazine as substrate.The optimum pH of the spore laccase was determined 7.0.When acetosyringone was added to the decolorizing system,its spore laccase could decolorize 93.0%,93.0%,79.0% and 92.5% of reactive black 5 in 4 hour,indigo carmine in 2 hour,reactive blue 19 and crystal violet in 6 hour.Conclusion] The spore laccase of strain CLb could decolorize commonly used dyes well in acetosyringonesystem.
