三叶木通中常春藤皂苷的提取工艺

目的]确定三叶木通中常春藤皂苷的最佳提取工艺条件。方法]比较了超声提取、索氏提取、沸水回流提取和常温浸提法时常春藤皂苷的得率,优选出超声提取方法,接着采用正交设计L9(34)方法,考察甲醇浓度、甲醇体积、超声时间、提取次数及酸解条件时的盐酸体积、水解时间、水解温度等因素对提取率的影响,采用HPLC法测定含量。结果]所考察的因素对三叶木通中常春藤皂苷提取的影响程度的顺序为提取时间(C)>甲醇浓度(A)>甲醇体积(B)>提取次数(D),水解时间对常春藤皂苷元得率影响明显。结论]三叶木通中常春藤皂苷的最佳提取工艺为:70%甲醇溶液80 ml超声提取90 min,水饱和正丁醇萃取3次,36%盐酸4 ml,70℃水浴回流水解4 h,氯仿萃取2次,合并氯仿层,旋转蒸干后甲醇溶解定容。

Extraction Process of Saponins PD in Akebia trifoliate

Objective] The extraction condition of saponins PD in Akebia trifoliate was studied.Method] Ultrasonic extraction,soxhlet extraction,heat reflux extraction and water extraction techniques were respectively used for extraction of saponins PD.According to the orthogonal design L9(34),the optimum ultrasonic extraction was sought through experimental investigation.The contents of saponins were determined by HPLC.Result] The order of factors to affect the saponins PD was ultrasonic action duration(C) > methanol concentration(A) > methanol volume(B) > extraction number of times(D).And hydrolysis time had an important influence on the extraction of saponins PD.Conclusion] The optimal conditions for extraction of saponins PD from Akebia trifoliate were as follows: 80 ml of 70% ethanol,90 min action duration,then extractived by n-butyl alcohol for 3 times,hydrolyzed with 36% hydrochloric acid for 4 h at 70 ℃,and extractived by chloroform.
