摘 要: | 目的]建立青刺果(Prinsepia utilis Royle)黄酮鉴别与含量测定的方法。方法]采用乙醇回流法从青刺果油粕中提取总黄酮,通过颜色反应和薄层层析对青刺果黄酮进行理化鉴别,并用Al(NO3)3-NaCO2络合比色法测定其含量。结果]供试品溶液的最大吸收波长为510nm,在15min内稳定。芦丁对照品溶液浓度(x)与吸光度值(y)的线性回归方程式:y=-2.571×10-3+13.163x,r=0.9998,线性关系良好;加样回收率在98.00%~104.00%,平均为101.16%,RSD=2.15。测得青刺果粗品中总黄酮含量为7.20%~7.30%,平均为7.25%±0.05%,RSD=0.69%;青刺果油粕中总黄酮含量为0.37%~0.38%,平均为0.38%±0.01%,RSD=1.54%;总黄酮得率为5.17%~5.25%,平均为5.20%±0.04%,RSD=0.80%。结论]该研究建立的紫外分光光度法简便、快速、准确、可靠,可用于青刺果总黄酮含量的测定。

Abstract: | Objective]The aim was to establish the methods for the identification and determination of flavonoids components in Prinsepia utilis Royle.Methods]The ethanol reflux extraction method was adopted to extract total flavonoids frorm Prinsepia utilis Royle,and then Color reaction and thin-layer chromatography(TCL) were used for physic-chemical identification of flavonoids,whose content was later determined by Al(NO3) 3-NaCO2 complexation colorimetry.Results]The maximum absorption wavelength of sample solution is 510 nm,which becomes stable within 15 min.There is a good linear relationship between the concentration of rutin control solution (x) and absorbance value (y) with the linear regression equation y=-2.571×103+13.163x,r=0.999 8; the average recovery rate is among the range of 98.00%-104.00% with an average of 101.16% ,RSD=2.15% .It is detected that the flavonoids content in Prinsepia utilis Royle is 7.20%-7.30% with an average of 7.25%±0.05% ,RSD=0.69% ; the flavonoids content in oil meal of Prinsepia utilis Royle is 0.37%-0.38% with a an average of 0.38%±0.01% ,RSD=1.54% ; The yield of flavonoids is 5.17%-5.25% with an average of 5.20%±0.04% ,RSD=0.80% .Conclusion]The method was simple,rapid,accurate and reliable,which could be applied to the determination of flavonoids in prinsepia utilis royle. |
