草菇gpd启动子的克隆及表达载体的构建

从草菇基因组中克隆内源强启动子，为草菇基因工程育种提供启动子元件材料。［方法］采用PCR技术从草菇基因组DNA中克隆gpd启动子片段，利用生物信息学手段对其序列特征和调控元件进行分析，并构建表达载体。［结果］从草菇基因组中克隆出2条特异gpd启动子片段，大小分别为1056和614bp；成功构建了分别以这2条启动子片段驱动的、以gfp基因为报告基因的2个表达载体。［结论］该研究为进一步利用基因工程手段培育品质优良的草菇新菌株奠定了基础。

Cloning of gpd Promoter from Volvariella volvacea and Constructing of Expression Vector
