摘 要: | 目的]获得制备丝素肽的简便高效的方法.方法]以废蚕丝为原料,用0.5%碳酸钠水溶液进行脱胶,硫酸进行水解,氧化钙水溶液中和沉淀,活性炭脱色制备丝素肽;采用正交试验L9(34)优化硫酸水解工艺条件.结果]试验表明,蚕丝蛋白在固液比1∶40 g/ml,硫酸浓度30%,水解温度80℃,水解时间9h的条件下进行水解时,丝素肽的得率最高.结论]研究可为丝素肽的进一步工业化生产提供可靠的理论依据.

Abstract: | Objective] To obtain simple and high efficient method for preparation of silk fibroin peptide.Method] With waste silk as raw materials,silk fibroin peptide was prepared by refining waste silk with 0.5% sodium carbonate solution,hydrolyzing silk fibroin with sulfuric acid,desalting with precipitation separation method,decolorizing with activated carbon.Hydrolysis process condition was optimized by using orthogonal test L9 (34).Result] The results showed that silk fibroin peptide yield was the highest when silk protein was hydrolyzed under the conditions of solid-liquid ratio of 1∶ 40,the sulfuric acid concentration of 40%,hydrolysis temperature of 80 ℃,hydrolysis time 9 h.Conclusion] This study provided reliable theoretical basis for the industrial production of silk fibroin peptide. |
