畜禽免疫增强剂中复合多糖的提取及免疫活性研究

目的]为中药有效成分的进一步研究提供理论依据。方法]通过正交试验对中药方剂中复合多糖的提取工艺进行筛选。以环磷酰胺作为免疫抑制剂建立小鼠免疫抑制模型,进行免疫脏器指数的检测、碳粒廓清试验、血清溶血素形成和外周血T淋巴细胞数检测,探讨中药复合多糖对免疫功能低下小鼠的影响。结果]中药方剂中复合多糖的最优提取条件为:温度60℃,料液比1∶40,超声处理30 min。中药复合多糖能明显提高正常小鼠脾脏指数、胸腺指数及免疫抑制小鼠脾脏指数;增强正常小鼠和免疫抑制小鼠单核巨噬细胞系统的吞噬功能;促进溶血素的形成并提高外周血中T淋巴细胞数。结论]该中药复合多糖对免疫抑制小鼠有免疫兴奋作用。

Research on the Isolation and Immunological Activity of the Compound Polysaccharide in Traditional Chinese Medical Herb

Objective] The theory reference basis for the research on the effective component in Chinese medical herb was provided through the experiment.Method]The experiment in the isolation of the compound polysaccharides from traditional Chinese medical herbs(HCP) was conducted and the best combination of isolation conditions was obtained through the orthogonal experiment.The model of mice was established with cyclophosphamide treatment and a series of tests were carried out,including the index of immune organs,the trial in carbon expurgation test,the formation of blood serum hemolysin and the observation of the number of T-lymphocyte population of peripheral blood.Results] The optimal extraction condition of compound polysaccharide was: the temperature,60 ℃;the ration of material to solution,1∶40 and the ultrasound treatment,30 min.The results showed that the Chinese herbal medicine compound polysaccharide markedly strengthened the immunity function by the improvement of the spleen and thymus index of normal mice and spleen index of immune suppressive mice;strengthened the phagocytosis of mononuclear phagocyte system and the number of T-lymphocytes in peripheral blood and stimulated the production of the hemolysin of normal and immune suppressive mice.Conclusion] The compound polysaccharide in Chinese herbal medicine can obviously improve the immunity function of mice.
