摘    要:以银杏叶片为材料,分别采用CTAB法、SDS法和TRIzol法提取银杏叶片总RNA.结果表明:CTAB法、SDS法所提取的RNA经电泳检测,28S rRNA、18S rRNA和5S rRNA 3务带清晰,RT-PCR得到的条带与目的片断大小一致,说明所提取的RNA纯度高、质量好;TRIzol法所得到的RNA降解严重,没有得到完整的电泳条带.CTAB法和SDS法适于富含多糖和多酚等物质的植物总RNA提取.

Abstract:In order to obtain high quality RNA from Ginkgo biloba L.leaf in which polyphenols and polysaccharides are rich,the method of CTAB,SDS and TRIzol were used to extract RNA and the effects of RNA extraction were studied with different methods.The results showed that three bands of 28S rRNA,18S rRNA and 5S rRNA were appeared after gel electrophoresis with the method of CTAB and SDS.The RNA samples were used to amplify cDNA through RT-PCR and the amplifications were observed in agarose gel by electrophoresis.The experimental results showed that the purity and concentration of the extracted RNA can meet the demands of molecular biology analysis.The RNA samples extracted with the method of TRIzol were degraded gravely and obtained without gel electrophoresis band.The method of CTAB and SDS are suited to extract the RNA of the plant tissues in which polyphenols and polysaccharides are rich.
