激素对也门铁叶片愈伤组织的诱导效果

目的]寻求诱导也门铁叶片愈伤组织的最佳激素配方。方法]以也门铁无菌苗叶片为外植体,以MS为基本培养基,分别添加不同浓度的2,4-D、BA和NAA,配制成24种激素组合,通过离体组织培养,探讨不同激素配比对也门铁叶片愈伤组织的诱导效果。结果]当NAA为0 mg/L,2,4-D与BA 2种激素以不同浓度组合,在2,4-D浓度是BA浓度的1.0~1.5倍时,也门铁愈伤组织诱导效率较高,但质量较差。当2,4-D为0 mg/L时,BA与NAA 2种激素组合的诱导率为0。当2,4-D、NAA与BA 3种激素组合且2,4-D与NAA的浓度比值为1时,也门铁愈伤组织的诱导时间短,愈伤组织质量较好。也门铁叶片愈伤组织诱导的最佳培养基配方是MS＋0.5 mg/L2,4-D＋0.5 mg/L NAA＋1.0 mg/L BA,诱导率为44.4%。结论]该研究为探索高效的也门铁组培快繁新方法提供科学依据。
