高良姜中黄酮类化合物的提取及精制研究

摘    要:目的]探索提取高良姜中黄酮类化合物的最佳方法。方法]通过单因素试验及正交试验确定乙醇浸提高良姜中黄酮类化合物的最佳提取条件,并利用双水相体系萃取精制黄酮类化合物。结果]高良姜中黄酮类化合物提取工艺的最佳单因素水平为提取温度70℃,溶剂浓度40%,固液比1∶300,提取时间4 h。正交试验结果显示影响高良姜中黄酮类化合物提取的各因素的顺序为固液比>温度>提取时间>酶用量>乙醇浓度。最佳提取条件为固液比1∶450,温度80℃,提取时间5 h,酶用量3%,乙醇浓度50%。最佳双水相萃取条件为:PEG浓度24%,硫酸铵浓度12%,温度25℃,pH值7.00。结论]试验结果可为高良姜中黄酮类化合物的提取分离及纯化提供技术参数。

Study on Extraction and Refining of Flavonoids from Alpinia officinarum Hance
Abstract:Objective] The aim was to explore the optimum method for the extraction of flavonoids from Alpinia officinarum Hance.Method]The optimum extraction condition for flavonoids from A.officinarum extracted by ethanol was determined by single factor experiment and orthogonal experiment,and the flavonoids were refined by aqueous two-phase system.Result] The optimum single factor level of the extraction technology for flavonoids from A.officinarum were as follows: the extraction temperature of 70 ℃,the concn.of solvent of 40%,the ratio of solid to liquid of 1∶300 and the extraction time of 4 h.The result of orthogonal experiment showed that the order of the factors affecting the extraction of flavonoids from A.officinarum was ratio of solid to liquid>temperature>extraction time>enzyme concn.>ethanol concn.The optimum extraction condition were the ratio of solid to liquid of 1∶450,the temperature of 80 ℃,the extraction time of 5 h,the enzyme concn.of 3% and the ethanol concn.of 50%.The optimum aqueous two-phase extraction condition were the PEG concn.of 24%,the ammonium sulfate concn.of 12%,the temperature of 25 ℃ and the pH value of 7.00.Conclusion] The test results could provide technical parameters for the extraction,separation and purification of flavonoids from A.officinarum.
