葛根总黄酮的提取及其抗氧化性能研究

摘    要:目的]优化提取葛根总黄酮的最佳工艺条件,研究其抗氧化性能。方法]用乙醇提取葛根总黄酮,通过正交试验,研究影响葛根总黄酮提取率的因素,确定最佳提取工艺参数;用硅胶柱层析法纯化葛根总黄酮提取物,采用BPCL法测定葛根总黄酮的抗氧化性能。结果]正交实验表明,影响葛根总黄酮提取率的4个因素的强弱顺序为:温度>乙醇浓度>固液比>提取次数。提取葛根总黄酮的最佳工艺条件为:乙醇浓度70%,固液比为1∶8,提取次数3次,每次3h,温度80℃,葛根总黄酮的提取率为22.9%,样品纯度为15.5%。结论]利用BPCL法测定葛根总黄酮对超氧自由基(O2-·)和羟基自由基(·OH)的抑制率,结果证明葛根总黄酮具有一定的抗氧化作用。

Abstract:Objective] The aim was to optimize the best technical conditions of extracting total flavone from Pueraria lobata,and studied its antioxidation property.Method] The total flavone from P.lobata was extracted with ethanol,the factors that influenced the extraction rate of total flavone from P.lobata was studied through orthogonal experiment,and the optimal extracting process parameters were identified.The extract of total flavone from P.lobata was purified by using silica gel column chromatography,and the antioxidation property of P.lobata total flavone was determined with BPCL method.Result] The orthogonal experiment showed that the sequence of four factors that affected the extraction rate of total flavone from P.lobata was:temperature > ethanol concentration > solid-liquid ratio > extracting times.The optimal technical conditions of extracting total flavone from P.lobata were:ethanol concn.of 70%,solid-liquid ratio of 1∶8,extracting at 80 ℃ for times with 3 h per time,under which,the extraction rate of total flavone from P.lobata was 22.9%,with the sample purity of 15.5%.Conclusion] The BPCL method was utilized to determine the inhibitory rate of total flavone from P.lobata to superoxide free radical(O2-·),and hydroxyl radical(·OH),and the result demonstrated that the total flavone from P.lobata had a certain antioxidant effects.
