酒精耐受型酿酒酵母菌的诱变筛选

摘    要:目的]筛选酒精耐受型酿酒酵母菌最佳菌株。方法]利用紫外线对酿酒酵母JL2008进行诱变处理,并结合氯化三苯四氮唑(TTC)进行筛选,对5株对酒精具有较强耐受性的突变株SC1~SC5的酒精耐受性进行比较,选择出2株酒精耐受性较强的突变株(SC1和SC5)进行酒精发酵,测定其生长速率和发酵性能。结果]2株菌的酒精耐受能力远远高于出发菌株,均能在酒精体积分数为9%的培养基上生长;在含糖15%的培养基中,SC1的酒精发酵终浓度和发酵效率与原始菌株大致相同;在含糖30%的培养基中,SC1的酒精发酵终浓度和发酵效率分别是16.54%和85.2%,均高于原始菌株。结论]突变株SC1在含酒精培养基中的生长和在浓醪条件下的发酵效果优于JL2008,是一株极具潜力的应用于浓醪发酵的酵母菌株。
