摘    要:目的]探讨秃疮花提取物(DLE)对四氯化碳(CCl4)诱导的小鼠急性肝损伤的影响。方法]小鼠灌胃给予不同剂量的DLE,连续10d后腹腔注射CCl4造成急性肝损伤,16 h后测定血清谷丙转氨酶(ALT)、谷草转氨酶(AST)活性及肝组织匀浆中丙二醛(MDA)和抗氧化酶活性;同时进行肝组织病理学观察。结果]与模型组相比,不同剂量的DLE给药组血清ALT、AST活性显著降低,肝组织匀浆MDA含量显著减少;抗氧化酶SOD、GSH-Px活性显著升高;病理切片表明DLE作用后肝组织损伤不同程度地减轻,其中剂量为2.0 g/kg时,肝组织的损伤程度较轻。结论]DLE对CCl4诱导的小鼠急性肝损伤具有很好的保护作用。其作用机理可能与DLE抑制抗氧化酶活性降低和抗自由基活性有关。

Abstract:Objective] The research aimed to discuss the effects of DLE on the acute liver injury in mice induced by CCl4.Method] Different dosage of DLE was injected through stomach in mice and CCl4 was given though intraperitoneal injection for 10 d continuously to cause the acute liver injury.After 16 hours,the activities of ALT and AST in the serum,and MDA content and antioxidant enzyme activity in the liver homogenate were determined.At the same time,the pathological observation of liver tissues was made.Result] Compared with model group, the activities of serum AST and ALT and MDA content in the liver homogenate in drug group with different dosages of DLE increased significantly.And the activities of antioxidant enzymes SOD and GSH-Px increased significantly.The pathological sections showed that liver injury was reduced to different extent after DLE treatment.When the dosage of DLE was 2.0 g/kg,the injury degree of liver tissues was slighter.Conclusion] DLE has good protective effect on the acute liver injury in mice induced by CCl4.Its mechanism might be related with the inhibition of DLE on the decrease of antioxidant enzyme activities and the anti-radicals activity.
