产红色素果蔬内生菌菌种鉴定

摘    要:目的]鉴定来自果蔬中的产红色素的内生菌菌株。方法]以来源于果蔬中的内生菌株为试验材料,对其进行生理生化特性分析及形态学和分子生物学鉴定。结果]该菌细胞卵形,单极芽殖;在液体培养基中有沉淀且有菌环;在固体培养基中,菌落红色,表面平滑、暗淡,边缘整齐,质地粘稠;无子囊孢子;无假菌丝形成。葡萄糖发酵为阴性;能同化硝酸钾;尿素分解呈阳性;明胶液化呈阴性;37℃呈阳性生长;DNA纯度较纯,核酸浓度为2 899 ng/ul。该酵母菌的18 S rDNA序列与粘性红圆酵母同源性达99.9%。结论]形态特征及生理生化和分子鉴定结果显示该菌为粘性红圆酵母。

Abstract:Objective] The aim was to identify the endophyte strain producing red pigment from fruit and vegetable.Method] With the endophyte strain from fruit and vegetable as tested material,its physiological and biochemical characteristics were analyzed and its morphology and molecular biology were identified.Result] The cell of the fungi was oval and monopole budding.The fungi had deposition and zone in liquid medium,its colony was red with smooth and gray surface,regular edge and sticky texture in solid medium,and the fungi had no ascospore and could not produce pseudohypha.The fungi was negative through glucose ferment and it could assimilate potassium nitrate.It showed positive through urea hydrolysis and negative through gelatin liquefaction.It also showed positive growth at 37 ℃.Its DNA purity was pure and its nucleic acid concn.was 2 899 ng/ul. The homology of the 18 S rDNA sequence from the yeast was 99.9% with that of the Rhodotorula mucilaginosa.Conclusion]The results of morphologic characteristics,physiological and biochemical and molecular identification showed that the fungi was R.mucilaginosa.
