摘    要:目的]对从Bacillus sp.SN02004-01菌所产高温海藻糖合酶产酶特性进行研究。方法]通过试验研究了菌株细胞的破壁处理工艺及所得细胞海藻糖合酶性质。结果]以2%甲苯+0.2%EDTA为破壁试剂,菌体浓度0.05g/ml,温度为35℃,150r/min处理3.0h为最佳处理条件;该细胞酶最适反应pH值7.0,最适反应温度为60℃;Cu2+、Zn2+对该酶有抑制作用。结论]在最佳处理条件下获得最高酶活为54.05u/ml,提高了2倍,说明该酶具有很好的生物反应特性。

Abstract:Objective]The research aimed to study the characterizations of thermostable trehalose synthease which produced by the bacterium Bacillus sp.SN02004-01.Method]The disrupted conditions of bacterium cells and characterizations of the thermostable trehalose synthase were studied by experiments.Result]The best disrupted conditions for bacterium cells treatment were 2% toluene and 0.2%EDTA as reagents,0.05 g/ml cell concentration,150 r/min oscillation 3 hours at 35 ℃.The pH and temperature of exhibited optimum activity for the thermostable trehalose synthase were 7.0 and 60℃.The thermostable trehalose synthase activity was restrained for Cu2+ and Zn2+.Conclusion]The maximal enzyme activity was 54.05 u/ml for the treatment of the optimum conditions and increased by 2 times.The thermostable trehalose synthetase had the fine biological response features.
