摘    要:目的]为甘薯品种的基因转化改良奠定基础。方法]以郑9728-4甘薯苗的枝条为材料,探讨不同激素组合对其愈伤组织产生、分化、增殖及生根的影响。结果]以甘薯顶芽下3~4节为外植体诱导无菌芽效果较好,外植体以0.1%的HgCl2灭菌6~7min效果较好;甘薯枝条腋芽诱导的最佳培养基为MS+BA1.0mg/L+NAA0.1mg/L;无菌芽各种外植体愈伤组织形成、分化和增殖的最佳培养基为BA1.0mg/L+IAA0.1mg/L(成愈率为98.7%,分化率为91.2%,增殖倍数为4.5倍,且丛芽生长健壮);再生芽生根的最佳培养基为1/2MS+NAA1.0mg/L。结论]建立了甘薯的高效再生体系。

Establishment of Efficient Regeneration System of Ipomoea batatas

Abstract:Objective] The aim was to lay a foundation for gene transformation and improvement of Ipomoea batatas varieties. Method] With branches of Zheng 9728-4 seedlings as materials,the effects of different hormone combinations on the generation,differentiation,proliferation and rooting of callus were discussed. Result] With 3rd-4th segments below top bud of I. batatas as explants,the effect of inducing aseptic buds was better. The effect of disinfection with 0.1% bichloride for 6-7 min on explants was better. The optimum medium for axillary bud induction from I. batatas branches was MS+BA 1.0 mg/L+NAA 0.1 mg/L. The optimum medium for generation,differentiation and proliferation of callus induced from aseptic buds of various explants was BA1.0 mg/L +IAA 0.1 mg/L,its callus formation rate,differentiation rate and proliferation times were 98.7%,91.2% and 4.5 times and the clustered buds grew healthy. The optimum medium for rooting of regenerated buds was 1/2MS+NAA 1.0 mg/L. Conclusion] The efficient regeneration system of I. batatas was established.
