龙椒2号大甜椒子叶离体培养中诱导不定芽的研究

摘    要:目的]采用子叶离体培养方法快速繁殖龙椒2号大甜椒,尤其在诱导不定芽阶段能更快、更多地诱导出不定芽。方法]在诱导不定芽阶段中以MS为基本培养基,添加不同的激素配比进行对比,以优选出最佳的激素配比方案。结果]生长素NAA对形成愈伤组织有主导作用,添加量以0.6 mg/L较为合适,6-BA和KT对促进芽分化都能起主导作用,6-BA以0.4 mg/L为宜,KT以1.0 mg/L为宜。结论]选用子叶切片转入MS+NAA0.6 mg/L培养基上,诱导出愈伤组织,然后将愈伤组织切块转入MS+6-BA0.4 mg/L+NAA0.5mg/L或MS+KT1.0 mg/L+NAA0.5 mg/L培养基上,分化出多数量的不定芽,然后利用这些不定芽进一步扩繁、生根,进而形成再生植株。

Study on the Inducing Adventitious Buds in Cotyledon Culture of Longjiao 2 Big Sweet Peppers in vitro

Abstract:Objective] The research aimed to reproduce Longjiao 2 Big Sweet Peppers rapidly by using the method of cotyledon culture in vitro,especially to faster induce more adventitious buds in the stage of inducing adventitious buds.Method] In the stage of inducing adventitious buds,taking MS as the basic medium and adding different hormone combination,the best hormone combination program was optimized.Result] NAA had the leading role in callus forming,and 0.6 mg/L NAA amount was more appropriate.6-BA and KT both could have the leading role in promoting bud differentiation,0.4 mg/L 6-BA amount and 1.0 mg/L KT amount were more appropriate.Conclusion] The cotyledon pieces were seeded into the medium of MS+6-BA 0.6 mg/L to induce the callus.Later callus were cut into pieces and which were seeded into the medium of MS+6-BA 0.4 mg/L+NAA 0.5 mg/L or the medium of MS+KT 1.0 mg/L+NAA 0.5 mg/L.When more numbers of adventitious buds were grown,those were used to do further propagation,rooting,and then become regenerated plants.
