小麦远缘杂交后代的高分子量麦谷蛋白亚基组成分析

摘    要:目的]为杂种后代的选择和品质育种提供参考依据。方法]以P5和郑州9023远缘杂交F2代株系为试材,用十二烷基磺酸钠-聚丙烯酰胺凝胶电泳法鉴定分析了杂交后代的高分子量麦谷蛋白亚基组成。结果]在Glu-A1,Glu-B1和Glu-D1 3个位点上分别均检测到2种亚基类型,3位点结合共出现2种组成类型。其中,在Glu-A1位点上,1种亚基出现频率最高,为66.7%;在Glu-B1位点上出现了7和7+8亚基,后者所占的比例是66.7%;Glu-D1位点上出现了2+12亚基和被世界公认的优质亚基5+10,其中,5+10亚基出现频率高达66.7%。杂交后代平均品质评分高达8分,两个株系的品质评分达到了10分。结论]该杂交获得的这些携带优良亚基的材料可以作为优质面包小麦新品种或育种材料使用,是栽培小麦品质改良的极好新种质资源。

Abstract:Objective] The purpose was to supply reference basis for the selection and quality breeding of hybridized progenies.Method] With lines of F2 hybrid from distant hybridization of P5 and Zhengzhou 9023 as tested materials,the subunit composition of high molecular weight glutenin in the hybridized progenies was identified and analyzed by sodium dodecyl sulfonate-polyacrylamide gel electrophoresis method.Result] Two subunit types were detected on 3 sites of Glu-A1,Glu-B1 and Glu-D1 resp.and 2 combination types appeared in the combinations of the 3 sites.Among them,on Glu-A1 site,the occurrence frequency of a subunit was highest,being 66.7%.The subunits 7 and 7 8 appeared on Glu-B1 site and the latter accounted for 66.7%.The subunits 2 12 and 5 10 that was generally acknowledged as good-quality subunit by the world appeared on Glu-D1 site,among them the occurrence frequency of the subunit 5 10 was up to 66.7%.The average quality score of hybridized progenies was up to 810 marks.Conclusion] These materials with good subunits obtained in the hybridization could be used as new wheat variety of good-quality bread or its breeding material.They were very good new germplasm resources for improving the quality of cultivated wheat.
