摘    要:目的]研究芥菜Cry2基因RNAi载体的构建与遗传转化,为后期对两者关系的进一步研究提供参考。方法]在克隆芥菜类茎瘤芥Cry2基因的基础上,构建该基因的RNAi载体,并导入根瘤农杆菌GV3103,获得工程菌株后,通过农杆菌浸染茎瘤芥子叶外植体进行植物体转化,并验证转化结果。结果]成功构建了茎瘤芥Cry2基因的RNAi载体,并建立了茎瘤芥遗传转化体系,获得了转基因植株。结论]茎瘤芥Cry2 RNAi的构建及遗传转化体系的建立为后期深入研究茎瘤芥熟期相关基因的功能及基因工程应用奠定了基础。

Construction of RNAi Vector of Cryptochrome 2 Gene and the Transformation of Brassica juncea

Abstract:Objective]Cryptochrome gene is one of the key genes in the light signal pathway which have important impacts on plant flowering.The Cryptochrome control seedling de-etiolation,dependent inhibition of Hypocotyl elongation,control of nutrition and anthocyanin accumulation,and the maintenance of plant endogenous rhythms in plants.research mustard cry2 gene RNAi carrier construction and genetic transformation Based on this study.Method]The research constructed the RNAi vector of cryptochrome 2 gene based on cloning of the gene from Tumorous stem mustard.Result]Then we successfully transfered it into agrobacterium tumefaciens strain GV3101 to form engineering strain.Dipped into plant callus through Agrobactrium tumefaciens,acquire transgenic plants.Conclusion]The construction of cry2 RNAi and the establishing of genetic transformation system set fundation for the research of the funtion of related gene in Tumorous stem mustard and the appllication of genetic engineering.
