摘    要:目的]研究围生期甲亢母鼠心脏中甲状腺激素受体在mRNA水平的差异表达。方法]首先建立妊娠合并甲亢模型,取围生期(孕16 d、孕18 d、孕20 d、产后5 d、产后10 d)母鼠心脏组织,提取总RNA。采用实时定量PCR方法,检测TRα1、TRβ1的表达量变化。结果]与正常母鼠相比,在围生期甲亢大鼠心脏中TRα1表达量在孕16 d上调213%,孕18 d上调64%,产后5 d上调888%,产后10 d上调200%。除产后10 d外,TRβ1变化不显著。结论]甲亢可以引起围生期大鼠TRα1在mRNA水平的表达量上调,TRα1的表达量与心功能的维持密切相关。

Differential Expression of Thyroid Hormone Receptors in the Heart of Hyperthyroid Rats in Perinatal Stage

Abstract:Objective] The research aimed to study the differential expression of thyroid hormone receptors in the heart of hyperthyroid rats in perinatal stage at mRNA level.Method] The pregnancy model with hyperthyroidism was first set up.The heart tissues of female rats in perinatal stage(the 16th,18th,20th day after gestation and the 5th day,10th day after parturition) were picked to extract total RNA.Using real-time quantitative PCR,the expression changes of thyroid hormone receptors(TRα1 and TRβ1) were detected.Result] Compared with normal female rats,the expression quantity of TRα1 in the heart of hyperthyroid rats in perinatal stage increased 213% on the 16th day after gestation,increased 64% on the 18th day after gestation,increased 888% on the 5th day after parturition and increased 200% on the 10th day after parturition.Except the expression quantity on the 10th day after parturition,the expression quantity of TRβ1 had no significant change.Conclusion] Hyperthyroidism could cause the up-regulation of the expression quantity of TRα1 in rats in perinatal stage at mRNA level.The expression quantity of TRα1 was closely related with the maintenance of cardiac function.
