超声集成双水相提取一枝蒿中总生物碱及抗氧化活性

摘    要:目的]优化超声提取法和双水相体系相结合提取一枝蒿总生物碱的工艺,对一枝蒿总生物碱抗氧化活性进行研究.方法]采用丙醇-硫酸铵双水相体系与超声波集成提取一枝蒿中总生物碱,以酸性染料比色法测定含量;通过考察对·OH清除和O2-·抑制能力来评价一枝蒿总生物碱抗氧化能力.结果]一枝蒿总生物碱最佳的提取条件为:硫酸铵用量8 g,丙醇用量25 ml,醇水比0.60∶1条件下超声提取60 min;在此条件下,一枝蒿中的总生物碱平均提取量0.436 mg/g.当总生物碱浓度为200 μg/ml时,对·OH清除率达到67.65%,对O2-·抑制率达30.92%.结论]该方法建立了一枝蒿总生物碱提取的最佳工艺,发现总生物碱具有良好的抗氧化活性.

Abstract:Objective] To optimize the extraction process of total alkaloids from Artemisia rupestris by ultrasonic coupled with aqueous two phase system,to study antioxidant activity of total alkaloids.Methods] Total alkaloids was extracted from Artemisia rupestris using propanolaqueous ammonium sulfate two phase system coupled with ultrasonic and the concentration of total alkaloids was determined with acid dye colorimetric method.Antioxidant activities were evaluated by capacity of total alkaloids for clearing · OH and controlling O2-·.Results] The optimum condition was 8.0 g ammonium sulfate,25 ml propanol,0.60∶1 propanol/water ratio,60 min of extraction,0.436 mg/g extraction rate of total alkaloids.When the concentration of total alkaloids was 200 μg/ml,the scavenging rate of · OH was 67.65%,and the suppression ratio of O2-· was 30.92%.Conclusion] The optimum extraction process of total alkaloids from Artemisia rupestris was obtained and the total alkaloids have good antioxidant activity.
