产脂肪酶细菌的筛选及碳、氮源对其产酶活性的影响

摘    要:目的]为扩大脂肪酶的生产源提供基础资料。方法]从贵阳市附近的炼油厂、粮油市场、贵州师范大学食堂采集土壤样品,从中筛选高产脂肪酶的细菌菌株。结果]经过初筛、复筛、分离和鉴定,从78份土样中共筛选到24株脂肪酶产生菌,这些菌株发酵液的脂肪酶活性为1.0-13.2 U/m l,其中从炼油厂土样中筛选到的A7菌株的产脂肪酶活性最强。分别以不同碳源和不同氮源培养A7菌株,发现各碳源对其产酶活性的促进作用分别为蔗糖〉葡萄糖〉果糖〉可溶性淀粉〉甘露醇,各氮源对其产酶活性的促进作用分别牛肉膏〉蛋白胨〉尿素〉黄豆饼粉、酵母粉〉硝酸钠。结论]炼油厂土样中细菌的产酶活性较高,细菌发酵产脂肪酶的最佳碳源为蔗糖,最佳氮源为牛肉膏。
