四倍体盾叶薯蓣快速繁殖的初步研究

摘    要:对二倍体和四倍体盾叶薯蓣无根苗生根条件进行了对比研究。结果表明 :以改良的MS培养基 (MS基础上铵态氮减半 )为基础 ,四倍体盾叶薯蓣愈伤组织诱导及不定芽分化的最佳培养基为 :MS(改良 ) + 6 BA 0 .5mg/L + 2 ,4 D 2 .0mg/L和MS(改良 ) +6 BA 2 .0mg/L +KT 0 .5mg/L。MS(改良 ) +NAA 0 .2mg/L培养基结合茎基部带部分倒圆锥形愈伤组织的切法最适于四倍体无根苗生根 ;以珍珠岩代替琼脂作支持物 ,既大大降低了生产成本 ,又提高了移栽成活率。

Abstract:Contrasting researches on rapid propagation of diploid and tetraploid Dioscorea zingiberensis Wright were made. Based on the modified MS-medium(with only half amount of N-NH 4),6 BA?0.5?mg/L+2,4 D?2.0?mg/L and 6 BA?2.0?mg/L+KT?0.5?mg/L were best for callus inducement and indefinite bud differentiation. With turbination cutting and perlite instead of agar, we established a kind of low-cost and highly-surviving method of rapid propagation of Tetraploid Dioscorea zingiberensis Wright.
