摘    要:探讨了稀释液、清洁剂及冷冻程序对猪精液冷冻保存效果的影响。结果显示:①稀释液Ⅳ保存鲜精,精子活率明显低于其他组(P<0.05);但其作为冻精的解冻稀释液时,效果最佳(P<0.05),说明猪精液冷冻保存时可采用第Ⅰ~Ⅲ组稀释液,而解冻时采用第Ⅳ组较佳。②稀释液中于不同阶段添加SDS(Sodiumlaurylsulfate,十二烷基硫酸钠)或OEP(OrvusESPaste,清洁剂)均能显著提高冷冻精子活率(P<0.05),SDS与OEP对冻精活率的影响无显著差异(P>0.05)。③精液冷冻程序采用-70℃、液氮熏蒸与直接液氮熏蒸在对解冻后冻精精子活率上无显著差异(P>0.05)。添加咖啡因能有效提高冻精的解冻后活率。

Abstract:The experiment in the effects of extender, detergent and cooling protocols on freezing boar semen was carried out. Fresh sperm were diluted by extendersⅠ~Ⅳ and enclosed in 5 ml straw. After placed in liquid nitrogen vapor, frozen sperm was thawed or stored for 3 h. The motility of sperm was detected. The result was following:①the motility in extender Ⅳ was lowest in four groups(at thawing: 60.0 %,57.2 %,56.3 % and 50.6 %,P<0.05; at 3 h storage: 17.9 %, 15.1 %, 13.5 % and 9.3 %,P<0.05); however, the extender Ⅳ was better than other groups as thawing dilution(at 3 h storage: 4.2 %, 6.8 %, 12.1 %, 32.8 %, P<0.05).②when detergent(SDS or OEP) were added to extender, the motility was promoted, but there was no significantly different between SDS and OEP.③ freezing protocols of 2-step cooling-down method(-70 ,-110 ℃) and 1-step cooling method(-110 ℃) had no effect on the percentage of motility after thawing. The addition of caffeine could promote the motility of frozen-thawed sperm.
