摘    要:根尖压片技术是研究植物中期染色体数目和形态的一种重要方法。以2个大菊品种的根尖为材料,以典型中期分裂相的数目、中期染色体的分散程度、聚缩程度和中期染色体的清晰性为衡量制片效果优劣的标准,通过对比不同预处理药剂、取材时间、预处理温度、预处理时间长短以及解离时间条件下的制片效果,得出如下结论:当根长至1 cm时,在9:00~10:00取根,用饱和对二氯苯溶液在4℃条件下预处理4 h,然后用卡诺固定液固定22~24 h,用1 mol/L的HCl60℃下解离10 min,能够得到数量较多、染色体分散良好、染色体形态清晰以及染色体长度适中的中期分裂相。

Root-tip Squashing Method for the Karyotype Analysis of Chrysanthemum morifolium

Abstract:Root-tip squashing method is important for the karyotypical study of plants.Two selected representative large-flowered Chrysanthemum morifoliam varieties were obtained in different time,treated with different kinds of pretreatment agent,different temperature,hour and dissociating time.The number of the mitosis metaphase cells,the degree of treatment complications and the fact that chromosomes were distinct and countable.The best conditions are as follows:pretreating roots of 1 cm with the saturated p-dichlorobenzene for 4 h at 4 ℃,fixing them with Carnoy's for 22-24 h and dissociating with 1 mol/L HCl at 60 ℃ for 10 min.This way can obtain slides of metaphase chromosomes with a high quality and quatity.
