真姬菇子实体多糖提取条件正交试验研究

摘    要:目的]筛选真姬菇子实体多糖的最佳提取条件,为真姬菇多糖的开发利用提供依据。方法]用蒸馏水提取真姬菇子实体多糖,以浸提温度、浸提时间、浸提比和浸提次数为主要因素,采用L9(34)正交试验进行提取工艺的优化。用苯酚-硫酸法测定多糖含量后计算多糖得率。结果]影响真姬菇子实体多糖提取的主要因素由大到小依次为浸提温度>浸提比>浸提次数>浸提时间。正交试验表明,当浸提温度为90℃,浸提时间为2.0h,浸提比为1∶30,1次浸提时,多糖得率最高,达到4.8%。结论]通过正交试验及其验证试验确定的真姬菇子实体多搪的最佳提取条件为:浸提时间2.5h,浸提温度90℃,浸提比1∶30,浸提次数2次,在此条件下,真姬菇子实体多糖得率可达5.1%。

Abstract:Objective] The study was to screen out the optimum conditions for extracting polysaccharide from Hypsizigus marmoreus fruit body, and provide the theoretical basis for the development and utilization of polysaccharides from H. marmoreus.Method] The polysaccharides from H. marmoreus fruit body was extracted with distilled water. With extraction temperature, extraction time , extraction ratio, extraction times as main factors, the L9(34) orthogonal experiment was used for optimizing the extraction technology. The yield rate of polysaccharide was calculated after the polysaccharide content was detected by phenol-vitriol. Result] The main factors of effecting on the extraction of the polysaccharides from H. marmoreus fruit body from big to small was extraction temperature>extraction ratio>extraction times>extraction time. The results of the orthogonal test showed that the polysaccharides yield rate was highest (4.8% ) when the extraction temperature was 90 ℃, the extraction time was 2.0 h, the extraction ratio was 1∶30 and the extraction times was 1. Conclusion] The best conditions for extracting the polysaccharide from H. marmoreus fruit body through the orthogonal test and its verification test was as follows,the extraction time of 2.5 h, the extraction temperature of 90 ℃,the extraction ratio of 1∶30 and the extraction times of 2,and the yield of polysaccharides from H. marmoreus fruit body could achieve 5.1%.
