摘    要:目的]探讨不同激活方法对胞质内显微受精(ICSI)猪卵母细胞原核形成及其早期发育的影响。方法]以体外成熟44~48 h的猪卵母细胞为试材,通过用ICSI法对其进行受精,然后用不同激活方法激活,研究了不同激活方法对ICSI卵母细胞原核形成和早期胚胎发育的影响。结果]化学激活处理组的猪卵母细胞的原核形成率显著高于没有经过激活处理组,Ion(5 min)+6-DMAP(4 h)组的原核形成率为38.08%,Ion(5 min)+NCSU23(3 h)+6-DMAP(4 h)组的原核形成率为47.78%。各处理组的分裂率差异不显著,但2个激活处理组的分裂率较高。不经激活处理组的囊胚率为3.82%,Ion(5 min)+6-DMAP(4 h)组的囊胚率为15.49%,Ion(5 min)+NCSU23(3 h)+6-DMAP(4 h)组的囊胚率为10.70%。结论]2种激活方法的原核形成率和囊胚率差异不显著,都可用于ICSI猪卵母细胞的激活。

Abstract:Objective] The research aimed to discuss the influences of different activation methods on the pronucleus formation and early development of porcine oocyte fertilized by intracytoplasmic sperm injection(ICSI).Method] With porcine oocyte being matured for 44-48 h in vitro as tested materials,they were fertilized by ICSI and then activated by different activation methods to study the influences of different activation methods on their pronucleus formation and early embryonic development.Result] The pronucleus formation rate of porcine oocyte in the group treated by chemical activation was significantly higher than that in non-activated group,the pronucleus formation rate of group Ion(5 min) + 6-DMAP(4 h) was 38.08% and that of group Ion(5 min) + NCSU23(3 h) + 6-DMAP(4 h) was 47.78%.The difference of division rate among treatment groups was not significant,but the division rates of the two activated groups were higher.The blastocyst rate of non-activated group was 3.82%,that of group Ion(5min) + 6-DMAP(4 h) was 15.49% and that of group Ion(5 min) + NCSU23(3 h) + 6-DMAP(4 h) was 10.70%.Conclusion] The diference of pronucleus formation rate and blastocyst rate between the two activation methods was not significant and both of them could be used to activate porcine oocyte fertilized by ICSI.
