摘    要:目的]寻找操作简便、耗时短、成本低的水稻基因组DNA提取方法。方法]分别以水稻幼嫩黄化叶片和老叶片为材料,用8种方法提取其中的DNA,测定所提DNA的浓度和纯度,并对其进行PCR扩增和电泳检测,比较各方法的提取效果。结果]8种方法提取的DNA浓度分别为35.15、30.80、67.30、26.15、23.55、8.95、48.05、54.26μg/ml,方法③提取的DNA浓度最大,但PCR扩增效果较差;改进的SDS法(方法⑦、⑧)提取的DNA纯度较高,PCR扩增产物电泳条带较亮,但这2种方法操作程序复杂,成本较高,提取每份样品的成本分别为14、31元,远高于其他提取方法。结论]除方法③外,其余5种简化方法均能得到较高质量的水稻基因组DNA,且提取成本远低于传统SDS法。

Abstract:Objective] The aim was to find the extraction method on rice genomic DNA with simple operation,short time and low cost.Method] With etiolated and old leaves of rice as the materials,their DNA were extracted by 8 kinds of methods,the conn.and purity of extracted DNA were determined and the extraction effects of different methods were compared by PCR amplification and electrophoresis detection.Result] The DNA concn.extracted by 8 kinds were 35.15,30.80,67.30,26.15,23.55,8.95,48.05,54.26 μg/ml resp.DNA concn.extracted by method 3 was the highest but its expansion effect by PCR was worse.The purities of DNA extracted by improved SDS methods(method ⑦ and ⑧) was higher and the electrophoresis strip of their PCR amplified products were brighter.But the operating process of those two methods was complex with higher cost and their costs extracting per sample were 14 and 31 yuan resp.far higher than that of other methods.Conclusion] Except method ③,other 5 simplified methods all could obtain high quality of rice genomic DNA and their costs were far lower than that of additional SDS method.
