摘    要:目的]对筛选得到的1株产纤维素酶的优良菌株进行鉴定,并研究其酶学性质。方法]利用传统分类学方法、电子显微镜观察、18SrDNA和序列的测定以及系统发育分析对1株分离自腐败水葫芦中产纤维素酶真菌D1进行分类鉴定,并对其产酶性能进行了初步研究。结果]各种指标鉴定表明其为青霉属真菌斜卧青霉(Penicillium decumbens)。该菌株经发酵培养120h,产酶酶活达到最高;活性在pH值4~6内稳定;pH值为5时酶活最高,50℃处理1h活性稳定。结论]该研究为纤维素酶的分离纯化和稳定性研究提供了必要的参考。

Abstract:Objective] The study aimed to identify one cellulolytic fungus and study its cellulase characteristics. Method] By using the traditional taxonomic methods, electronic microscope examination, 18S rDNA, sequences and phylogenetic analysis, an cellulolytic fungus D1 isolated from the Water Hyacinth was identified and its cellulase characteristics was studied. Result] All kinds of identifications showed that the cellulolytic fungus was Penicillium decumbens.The metabolite showed strong cellulase activity after cultured for 120 hours. This activity remained stable at 50 ℃ for 1 h treatment and pH value 4-6. Conclusion] The study provided the reference for separation, purification and stability study of cellulase.These data suggested that proper methods for cellulase purification and stability study.
