摘    要:目的]从鲜牛奶中分离植物乳杆菌,为牛奶中植物乳杆菌的开发利用提供依据。方法]将鲜牛奶稀释,进行涂布转接,再通过活菌培养、形态特征观察、生理生化试验,结合16S rDNA序列分析进行鉴定。结果]从鲜牛奶中分离得到1株细菌(D14),菌体形态为杆状,单个或链状排列,无芽孢,菌落凸起,边缘光滑细腻,呈白色,直径0.4~2.2 mm,有酸味。再结合各生理生化试验和16S rDNA序列分析,将其鉴定为植物乳杆菌(Lactobacillus plantarum)。结论]从鲜牛奶中分离得到的植物乳杆菌D14来源安全,有望应用于食品和饲料发酵,具有一定的应用前景。

Isolation and Identification of Lactobacillus plantarum D14 in Fresh Milk

Abstract:Objective]The study aimed to isolate Lactobacillus plantarum from the fresh milk and provide the basis for the development and utilization of plant lactobacillus.Method]The fresh milk was diluted to conduct the coating connecting and then through living bacterium culture,morphology observation,physiological and biochemical tests,combining 16S rDNA sequence analysis,the bacterium was identified.Result]A strain of bacterium named as D14 was isolated from the fresh milk.The morphology of the bacteria was rod-shaped,individual or chain-shaped arrangement,no spores,colonies protuberant,edge smooth and fine,white,0.4-2.2 mm in diameter,with sour taste.Combined with the physiological and biochemical tests and 16S rDNA sequence analysis,the bacterium was identified as L.plantarum.Conclusion]The L.plantarum D14 isolated from the fresh milk was safe in the source,so it is expected to be used for the food and feed fermentation,which had certain application prospect.
