湖北省不同地区油茶遗传多样性的AFLP分析

目的]研究湖北省油茶资源的遗传多样性。方法]采用AFLP标记对湖北省5个地区油茶的遗传多样性进行研究。结果]用4对引物对48个个体进行扩增,获得清晰的扩增条带45条,其中多态性条带38条,占84.44%;油茶居群的多态位点在22.22%～53.33%,有效等位基因数（Ne）在1.140 1～1.228 3,Nei′s基因多样性指数在0.084 3～0.144 2,Shannon′s信息指数在0.126 2～0.227 8。聚类分析将居群分为2大类群,在分子水平上显示了其亲缘关系。结论]油茶不同居群间遗传多样性水平相差较大,油茶遗传分化主要存在于居群内。为今后油茶育种亲本的选配提供理论依据。

AFLP Analysis on the Genetic Diversity of Camellia oleifera from Different Regions of Hubei Province

Objective]To study the genetic diversity of Camellia oleifera in Hubei Province.Method]The genetic diversity of C.oleifea from five regions of Hubei Province was verified by AFLP marker.Result]A total number of 45 discernable fragments were amplified from the 48 samples with 4 primers,38 of which were polymorphic,and their polymorphism frequency was 84.44%;the percentage of polymorphic loci of C.oleifera populations ranged from 22.22% to 53.33%,effective number of alleles ranged from 1.140 1 to 1.228 3,Nei′s gene diversity ranged from 0.084 3 to 0.144 2,and Shannon′s information index ranged from 0.126 2 to 0.227 8.The tested populations were separated into two clusters,showing their parent relationship at the molecular level.Conclusion]Genetic diversity differed largely among different populations,and the genetic differentiation mainly occurred within populations.The genetic relationship revealed by AFLP would be useful for selecting the parents in Camellia oleifera breeding programs in future.
