摘    要:目的]探究拟硬刺高原鳅种群的遗传多样性和分化程度。方法]采用聚合酶链式反应(PCR)和直接测序方法获得拟硬刺高原鳅线粒体DNA(mtDNA)细胞色素b(Cytb)基因的全序列。结果]拟硬刺高原鳅3个种群共58个个体均获得Cytb基因的全长序列(1 140 bp),共定义了20个单倍型,拟硬刺高原鳅宝库河种群遗传多样性低于黄河种群。结论]拟硬刺高原鳅黄河种群和宝库河种群间存在显著的分化,建议在实践中将2种群分开管理和保护,以达到保护种群遗传多样性的目的。

Study on Genetic Diversity of Triplophysa pseudoscleroptera Based on Cyt b Gene Sequence

Abstract:Objective]The research aimed to investigate the genetic diversity and differentiation of Triplophysa pseudoscleroptera populations.Method]The complete sequences of the mitochondrial cytochrome b gene of Triplophysa pseudoscleroptera were obtained by using of PCR and DNA sequencing.Result]The complete sequences of Cyt b gene(1 140 bp) were obtained from 58 individuals of Triplophysa pseudoscleroptera,representing three populations from Yellow River and Baoku River,and a total of 20 haplotypes were identified.The genetic diversity of population from Baoku River was lower than that of population from Yellow River.Conclusion]The significant genetic differentiation between populations from Yellow River and Baoku River suggested that they should be managed and conserved separately and that efforts should be directed towards preserving the genetic diversity of each group.
