摘    要:目的]为进一步提高海洋菌Bacillus sphaericus M4产多糖的量,对其发酵产多糖的条件进行优化.方法]分别研究发酵温度、转速对发酵的影响,并通过两段法对Bacillus sphaericus M4产多糖的条件进行优化.结果]菌体最适生长温度为31℃,最佳转速为200r/min;两段法优化的结果表明:摇瓶转速由200 r/min降为100 r/min,多糖产量会由8.32 g/L提高至10.55 g/L;温度由31℃升至34℃C,多糖产量会由10.35 g/L提高至12.16 g/L.结论]为Bacillus sphaericus M4胞外多糖的工业化生产奠定了基础.

Two Paragraphs Optimization for Exopolysaccharide Production of Marine Bacteria M4 based on the Stress Effects

Abstract:Objective] To further improve the yield of polysaccharide,the fermentation conditions of marine bacteria Bacillus sphaericus M4 were optimized.Method] The effects of fermentation temperature and rotation speed on fermentation were studied,and the polysaccharide production conditions of Bacillus sphaericus M4 were optimized by two-stage method.Result] The optimum growth temperature of bacteria was 31 ℃,the optimum rotation speed was 200 r/min.The result of two-stage optimization method showed: when the rotation speed of shake flask was decreased from 200 r/min to 100 r/min,the yield of polysaccharide was increased from 8.32 g/L to 10.55 g/L;when the temperature was increased from 31 ℃ to 34 ℃,the yield of polysaccharide was also increased from 10.35 g/L to 12.16 g/L.Conclusion] It prepared the ground for the industrialized production of exopolysaccharide of Bacillus sphaericus M4.
